Llantwit Major have been drawn at home to Afan Lido in the first round of the Nathaniel MG Cup.
It will mark Karl Lewis' first game as manager in three years following his recent return to the Cymru South club.
The winners will get home advantage against either Swansea City or Carmarthen Town in the second round.
Llantwit will be aiming to go better their second round exit at home to Cymru Premier club Cardiff Met in last season's competition.
Swansea are one of just two wildcard entries in the league cup - with Cardiff City entertaining Ammanford in the first round.
The draw, announced on Wednesday night, also includes a tie between two newly-promoted clubs - Ynyshir Albions and Cardiff Draconians.
Barry Town United enter the fray in the second round, starting with a home game against Aberystwyth Town, who were relegated from the Cymru Premier after 33 years as one of the league's founder members.
Barry, under then-manager Steve Jenkins (now the club's head of football), reached the semi-finals last season before going out to eventual winners The New Saints, who went onto beat Aberystwyth.
The first round ties will be played on the weekend of Friday 18th - Sunday 20th July - with the second round being played on the weekend of Friday 1st - Sunday 3rd August.
