The B4270 was closed for around three hours after a trailer overturned between Cowbridge and Llandow.
Llantwit Major Road was shut both ways following the collision at around 9.10pm on Wednesday evening.
South Wales Police said there were no reports of any injuries and the road was reopened shortly after midnight.
